Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10–20 minutes followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is kidney tissue.

Target Information

Primary steroid-resistant nephrotic syndrome (SRNS) is characterized by childhood onset of proteinuria and progression to end-stage renal disease. Kidney podocytes and their slit diaphragms form the final barrier to urinary protein loss.
Congenital nephrotic syndrome (CNS) is caused by mutations in NPHS1 (nephrin) or NPHS2.
Nephrin is a cell adhesion molecule that plays a crucial role in maintaining the glomerular filtration barrier and anchoring the podocyte slit diaphragm to the actin cytoskeleton.
Mutations in the nephrin gene are reportedly associated with congenital nephrosis.
Nephrin also participates in anti-apoptotic activity in podocyte slit diaphragm, potentially through VEGF signaling.
