Product Specific Information

This H5C6 monoclonal antibody reacts with human CD63, a type III tetraspanin transmembrane protein. CD63 is expressed intracellularly on lysosomes, endosomes, and granules of resting platelets and basophils. Cell surface expression is detected on activated basophils, platelets, monocytes, macrophages, and granulocytes. It is also expressed on endothelial cells, fibroblasts, and smooth muscle cells. CD63 associates with integrins (VLA-3, VLA-6) and TIMP-1 to mediate allergic responses.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.5 µg) per test (final volume 100 µL). Cell number may range from 10⁵ to 10⁸ cells/test.

Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation: Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD63 (LAMP-3) is a glycoprotein of the tetraspanin family found in late endosomes, lysosomes, and secretory vesicles. It is also present in the plasma membrane following cell activation, serving as a basophil activation marker. CD63 interacts with integrins and affects phagocytosis, cell migration, and ion channel regulation. It functions as a T-cell costimulation molecule and can predict carcinoma prognosis. CD63 is expressed on activated platelets, monocytes, macrophages, and weakly on granulocytes, B, and T cells. It is identical to melanoma-associated antigen ME491 and platelet antigen PTLGP40. CD63 dysfunction is associated with melanoma and Hermansky-Pudlak Syndrome.
