Target Information

EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1)는 인간 염색체 7에 위치한 EGFR 유전자에 의해 암호화됩니다.
EGFR은 HER/ERbB 계열의 단백질로, ERbB2, ERbB3, ERbB4를 포함합니다.
리간드(EGF, TGFα 등)가 결합하면 EGFR은 이합체화 후 세포질 도메인 내 티로신 잔기들이 자가 인산화됩니다.
EGFR 활성화는 Ras-ERK, PI3K-Akt, Jak-STAT, PKC 경로를 포함한 다양한 신호 전달을 유도하여 세포 성장과 증식을 조절합니다.
Y1086 잔기 인산화는 GRB2 단백질 결합을 통해 MAPK 경로를 활성화합니다.
활성화 후 EGFR은 내포되어 분해 또는 재활용 경로로 이동합니다.
EGFR 유전자 변이는 폐암과 관련이 있으며, 여러 종양에서 EGFR 과발현은 불량한 예후와 연관됩니다.
