Target Information

ADRBK1 (GRK2) is a serine/threonine kinase. Many G protein-coupled receptors (GPCRs) are phosphorylated by G protein-dependent receptor kinases (GRKs).
GRK2 (83 kDa), one of six identified members, is ubiquitously expressed in mammals.
After ligand binding and interaction with heterotrimeric G proteins, GPCRs are phosphorylated by GRKs.
Internalization of GPCRs, regulated by beta-arrestin-1, activates the Ras–Raf–ERK1&2 signaling pathway.
GRK2 activity is tightly regulated by phosphorylation (PKC, Src, ERK1&2) and protein interactions.
ERK phosphorylates and inactivates GRK2 on serine 670 in a negative feedback mechanism.
