Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is tonsil tissue.

Target Information

CD7 (gp40, Leu9) is a 40 kDa member of the immunoglobulin gene superfamily.
The N-terminal amino acids (1–107) are highly homologous to Ig kappa-L chains, while the C-terminal extracellular region is proline-rich and may form a stalk projecting the Ig domain.
CD7 is expressed on most immature and mature T-lymphocytes and T cell leukemia cells, as well as on natural killer cells, a subset of normal B cells, and malignant B cells.
Cross-linking surface CD7 modulates T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation.
CD7 directly associates with phosphoinositol 3'-kinase, and its ligation induces production of D-3 phosphoinositides and tyrosine phosphorylation.
Expression of CD7 is an important diagnostic marker in leukemia.
