NOS (Nitric Oxide Synthase) activity requires homodimerization and utilizes three cosubstrates (L-arginine, NADPH, O₂) and five cofactors (FAD, FMN, calmodulin, tetrahydrobiopterin, heme).
Three distinct NOS isoforms exist, encoded by separate genes:
- NOS1 (nNOS): neuronal form, Ca²⁺/CaM-dependent
- NOS3 (eNOS): endothelial form, Ca²⁺/CaM-dependent
- NOS2 (iNOS): inducible form
