Product Specific Information

This antibody recognizes both mutant and wild-type mammalian p53 protein. It reacts with an epitope between amino acid residues 370 and 378 (near the C-terminus of the protein), corresponding to the sequence KKGQSTSRHK.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest.

The structure of p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 plays a vital role in preventing proliferation of damaged cells and acts as an anticancer agent. It is also linked to aging, oxidative stress, and cognitive function deficits when overexpressed.

Mutations in the TP53 gene are common in human cancers, leading to loss of tumor suppressor activity. Germline mutations can cause hereditary cancer syndromes such as Li-Fraumeni syndrome.
