This antibody detects endogenous levels of Tau only when phosphorylated at Ser519, a site historically referenced as Ser202.
Target Information
Tau is a neuronal microtubule-associated protein predominantly localized on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus associates with neural plasma membrane components, functioning as a linker between the two. Axonal polarity is determined by TAU/MAPT localization within the neuronal cell body domain defined by the centrosome.
Short isoforms contribute to cytoskeletal plasticity, whereas longer isoforms aid in stabilization. In its hyper-phosphorylated form, Tau constitutes paired helical filaments (PHF), the main component of neurofibrillary tangles in Alzheimer’s disease (AD). Hyper-phosphorylation disrupts Tau’s microtubule-binding ability, leading to microtubule destabilization and neuronal degeneration in AD brains.
Tau phosphorylation is mediated by several kinases, including GSK-3β, protein kinase A (PKA), cyclin-dependent kinase 5 (CDK5), and casein kinase II. Hyper-phosphorylated Tau is also observed in other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
