Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor functions as a ligand-activated transcription factor. Upon binding to estrogen, it undergoes a conformational change that enables dimerization and binding to estrogen response elements (ERE) in DNA. This interaction regulates gene transcription through coactivator or corepressor recruitment.

There are two main forms of the estrogen receptor, ERα (ESR1) and ERβ (ESR2). Alternative RNA splicing produces multiple ERα isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are expressed in many tissues, crucial for sexual development, reproduction, and bone physiology, and are implicated in diseases such as breast and endometrial cancers, and osteoporosis.
