Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction can regulate gene transcription through coactivator or corepressor recruitment.

There are two major forms of the estrogen receptor, alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing produces at least four known isoforms of ER-alpha (66, 53, 47, and 35 kDa). Estrogen receptors are widely expressed in various tissues, essential for sexual development and reproductive function, and also influence bone metabolism. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
