Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. ER regulates gene transcription through coactivator or corepressor recruitment.
Two main forms exist: ER alpha (ESR1) and ER beta (ESR2). Alternative RNA splicing produces at least four ER-alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). ERs are widely expressed in various tissues and are essential for sexual development, reproductive function, and bone physiology. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
