Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA. ER can regulate gene transcription positively or negatively via recruitment of coactivator or corepressor proteins.
There are two forms of estrogen receptor, alpha and beta, encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing produces at least four ER-alpha isoforms (66 kDa, 53 kDa, 47 kDa, and 35 kDa). Estrogen receptors are widely expressed across tissues, essential for sexual development and reproductive function, and implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
