Description:
Monoclonal antibody 54 recognizes mouse Tim-4 (T cell, Ig, mucin). Unlike other Tim family members, Tim-4 is not expressed on T lymphocytes but is found on antigen-presenting cells such as dendritic cells and macrophages. Tim-4 interacts with Tim-1, suggesting a role in Th1/Th2 regulation balance. It also acts as a receptor for phosphatidylserine, playing a role in the engulfment of apoptotic cells. Expression is found on a subset of CD11c⁺ splenic cells.

This 54 (RMT4-54) antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of resident peritoneal macrophages. Recommended ≤0.25 µg per test (based on 100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Target Information

The TIM (T cell immunoglobulin and mucin domain containing) family encodes cell surface receptors involved in the regulation of Th1 and Th2 cell-mediated immunity. TIM-4, preferentially expressed on macrophages and dendritic cells, is the natural ligand of TIM-1, leading to T-cell expansion and cytokine production. Unlike other TIM family members, TIM-4 lacks a tyrosine phosphorylation signal sequence in its intracellular domain. The TIM-4 gene maps to a locus associated with asthma predisposition in both mice and humans, and due to its connection with TIM-1-triggered Th2 responsiveness, it may be considered a candidate gene for asthma susceptibility.
