Product Specific Information
The eBioMLC-51 monoclonal antibody recognizes mouse Nanog, a homeobox transcription factor crucial for maintaining pluripotency in embryonic stem cells, independent of LIF/Stat3. Nanog is expressed in early embryos, ICM of blastocyst, embryonic stem (ES) cells, and embryonic germ (EG) cells. Its expression decreases upon differentiation, making it a key marker for stem cell pluripotency.
Nanog functions as a transcriptional activator with activation domains CD2 and WR in the C-terminus and one in the N-terminus. The CD2 domain is unique to Nanog, while the NK2 DNA-binding domain is conserved. Immunoblotting with eBioMLC-51 identifies a ~45 kDa band in F9 lysate, but not in NIH3T3 or mouse spleen lysates.
For intracellular staining of mouse Nanog, use the eBioscience Foxp3 Staining Buffer Set (Product #00-5523-00).
Applications reported include intracellular staining for flow cytometry and immunocytochemistry.

Target Information
NANOG (Nanog homeobox) is a homeodomain protein regulating pluripotency and differentiation in embryonic stem cells. NANOG mRNA is present in pluripotent mouse and human cell lines and absent in differentiated cells. Human NANOG shares 52% amino acid identity with mouse NANOG (85% in homeodomain).

NANOG expression is detected in the inner cell mass of blastocysts and is critical for ES cell self-renewal. Overexpression promotes S-phase entry and proliferation. It is also a key factor in generating induced pluripotent stem (iPS) cells alongside Oct4, Sox2, Klf4, Lin28, and c-Myc, offering potential applications in regenerative medicine.
