Target Information
TPA converts the abundant, but inactive, zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. It plays an essential role in tissue remodeling, degradation, and cell migration. TPA binds fibrin with high affinity, enhancing catalytic efficiency up to 1000-fold. It also interacts with heparin, laminin, fibronectin, mannose receptor, and LRP1, influencing plasminogen activation and clearance. Additional interactions with annexin II and cytokeratin 8 are known. Binding to vascular smooth muscle cells (VSMC) reduces TPA inhibition by PAI-1 significantly.
