Product Specific Information
- Purified from rabbit serum by sequential epitope-specific chromatography.
- Contains enough material for 10 mini-blots.
- Antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to phosphorylation sites to remove cross-reactivity with non-phosphorylated ERK1&2.
- Final product generated by affinity chromatography using ERK1&2-derived peptide phosphorylated at Thr202/185 and Tyr204/187.
- Validated for western blotting and immunostaining; other applications may require optimization.
- Positive controls: PC12 cells ± Sorbitol; NIH3T3 ± PDGF; HEK293 ± UV.
Target Information
ERK1 and ERK2 are key kinases involved in meiosis, mitosis, and postmitotic cellular functions. They are activated by diverse stimuli such as growth factors, cytokines, viral infection, and GPCR ligands. Upon activation, Raf phosphorylates MEK1/2, which in turn phosphorylates ERK1/2 at specific threonine and tyrosine residues. Activated ERKs regulate cytoplasmic targets or translocate to the nucleus to activate transcription factors like c-Fos and Elk-1.
