Target Information

The modification of proteins with ubiquitin is a key cellular mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves three classes of enzymes:

1. E1 ubiquitin-activating enzymes
2. E2 ubiquitin-conjugating enzymes
3. E3 ubiquitin-protein ligases

UBE3B encodes an E3 ubiquitin-conjugating enzyme that transfers ubiquitin from E2 enzymes to specific substrates. The longer isoform contains a HECT domain in its C-terminus, which serves as the catalytic site for ubiquitin transfer and interacts with E2 conjugases. Shorter isoforms lacking this domain are unlikely to bind E2 enzymes. Alternative splicing generates distinct isoforms of this gene.
