Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.
Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.4 µg)/test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL volume (10⁵–10⁸ cells/test).

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology:
Based on Phiton technology utilizing novel nucleic acid dye structures for engineered fluorescent signatures with reduced spillover and spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

The CD3 subunit complex is crucial in transducing antigen-recognition signals into T cells and regulating TCR surface expression.
CD3 subunits (gamma, delta, epsilon, zeta) are part of the immunoglobulin superfamily on chromosome 11.
They have long cytoplasmic tails that interact with signal transduction molecules via tyrosine-based motifs.
CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
