Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Recommended use: 5 µL (0.4 µg)/test in 100 µL final volume, with 10⁵–10⁸ cells/test.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) improves staining performance. Add 5 µL per sample before combining antibody cocktails with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures for optimized spectral performance and minimized spillover.

Target Information

CD3 complex transduces antigen-recognition signals and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These proteins associate with signal transduction molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
