Product Specific Information

Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking via UCHT1 initiates intracellular signaling leading to activation and proliferation.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended at 5 µL (0.4 µg)/test for a final volume of 100 µL. Cell number may range from 10⁵–10⁸ cells/test.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Buffer Recommendation:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample to antibody cocktails or single-color controls for optimal staining performance.

Technology:
NovaFluor conjugates are based on Phiton technology using nucleic acid dye structures engineered for optimized fluorescent signatures with reduced spillover and spread.

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are immunoglobulin superfamily members encoded on chromosome 11. They associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells. It is absent on B or NK cells. Mutations in CD3 genes can cause immunodeficiency.
