Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (gamma and delta), the epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.4 µg)/test.
A test is defined as the antibody amount (µg) used to stain a cell sample in 100 µL final volume. Cell number can range from 10⁵ to 10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton technology utilizing novel nucleic acid dye structures engineered for optimized fluorescence signatures with reduced spillover and spread impacts.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. Activation through TCR involves CD3 gamma, delta, epsilon, and zeta subunits—members of the immunoglobulin superfamily encoded on chromosome 11. These components have long cytoplasmic tails associated with signal transduction molecules via tyrosine-based motifs.

CD3 is expressed on:

  • 68–82% of normal peripheral blood lymphocytes
  • 65–85% of thymocytes
  • Purkinje cells in the cerebellum

Not expressed on B or NK cells. Reduced T lymphocyte percentages can occur in autoimmune diseases.
Defects in CD3 genes are associated with CD3 immunodeficiency.
