Target Information

Human immunodeficiency virus (HIV) and related viruses require coreceptors, in addition to CD4, to infect target cells. Several G protein-coupled receptors, including CCR5, CXCR4, CCR3, CCR2b, and CCR8, have been identified as HIV coreceptors. Among them, CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains, facilitating fusion and entry into human white blood cells.

CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Beta-chemokines such as RANTES, MIP-α, and MIP-β act as ligands for CCR5 and can prevent infection by M-tropic HIV-1. CCR5 interacts with the surface CD4-gp120 complex of HIV, leading to membrane fusion and viral entry. The amino-terminal domain and extracellular loops of CCR5 serve as binding sites for HIV. CCR5 mRNA is expressed in lymphoid organs and monocytes.
