Product Specific Information

The monoclonal antibody eBio440c recognizes Siglec-H, a protein exclusively found on plasmacytoid dendritic cells (pDCs) or type I IFN-producing cells (IPC) in naive mice.
Mouse IPCs are typically PDCA+, CD11c+, CD11b-, B220+, and Ly-6C+, and respond rapidly to viral infection.

Siglec-H is a transmembrane protein of the Ig superfamily, similar to CD33, that binds sialic acid but lacks a cytoplasmic ITIM domain. It associates with DAP12 to enable signal transduction.
The eBio440c antibody has been shown to inhibit pDC function by reducing IFNα secretion in response to CpG stimulation.
Some mouse strains (e.g., SJL) exhibit higher pDC percentages compared to C57BL/6.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Fluorophore Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass.
Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright conjugates to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57).
Performance after fixation should be verified empirically.
