Target Information

Several serine/threonine protein kinases are intermediates in signal transduction pathways, including ERK/MAP kinases, ribosomal S6 kinase (Rsk), and Raf-1.
Raf-1 is a cytoplasmic protein with intrinsic serine/threonine activity, broadly expressed in nearly all tested cell lines. It is the cellular homolog of v-Raf, the transforming gene product of the 3611 strain of murine sarcoma virus.
Unregulated kinase activity of v-Raf is associated with transformation and mitogenesis, while Raf-1 activity is normally suppressed by a regulatory N-terminal domain.
A-Raf, another member of the Raf gene family, shows substantial homology to Raf-1 within the kinase domain but less elsewhere. Expression of A-Raf is highest in urogenital tissues and kidney, and lowest in brain tissue.
