Description:
The IP26 monoclonal antibody reacts with the alpha/beta chain of human TCR, which is expressed by the majority of peripheral T cells.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.5 µg) per test.
A test is defined as the amount of antibody sufficient to stain a cell sample in 100 µL final volume.
Optimal cell number: 10⁵–10⁸ cells/test (empirically determined).
Note: Staining may be obstructed when co-stained with OKT3 (anti-human CD3) antibody.

Target Information

The T cell antigen receptor (TCR) is a ligand-specific alpha/beta heterodimer non-covalently associated with invariant CD3 subunits (γ, δ, ε, ζ). These complexes are essential for efficient surface expression and signal transduction during T cell activation, leading to differentiation, proliferation, and cytokine production.

Most human T cells express TCR α/β and either CD4 or CD8 (single positive). A small subset lacking both markers (double negative) can increase in autoimmune or immunodeficiency disorders. TCR γ/δ T cells, mainly found in epithelial tissues, show less diversity and distinct antigen recognition. TCR engagement by superantigens can cause toxic shock syndrome, and autoantibodies to TCR Vβ segments have been linked to autoimmune diseases such as RA and SLE.
