Product Specific Information

The 122 monoclonal antibody reacts with mouse B7-DC, also known as PD-L2. B7-DC is a member of the B7 family with a predicted molecular weight of approximately 25 kDa and a short cytoplasmic tail (4 amino acids). It is primarily expressed by subpopulations of dendritic cells and monocytes/macrophages in mice.

B7-DC acts as a ligand for PD-1, influencing T cell proliferation depending on activation state. Clone 122 has been shown to block binding of TY25, another mAb specific for mouse B7-DC.

Target Information

Programmed death-ligand 2 (PD-L2, B7-DC) is a B7 family member that binds PD-1, regulating immune responses. Expressed mainly in dendritic cells, PD-L2 can be induced in various cell types by cytokines such as IL-4, TNF-α, and IFN-γ. It has a higher affinity for PD-1 than PD-L1 and functions as an inhibitory immune checkpoint molecule.

PD-L2 can also interact with repulsive guidance molecule b (RGMb), influencing respiratory tolerance via BMP/neogenin signaling. The human PD-L2 monomer consists of a 201-aa extracellular domain, a 21-aa transmembrane domain, and a 32-aa cytoplasmic domain.
