Product Specific Information

The A20 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.1-expressing mouse strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed by all leukocytes in these strains.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes, ≤0.5 µg/test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

NovaFluor Compatibility:
Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, add 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
Excitation: 509 nm; Emission: 614 nm; Laser: 488 nm (Blue).

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases and is essential for lymphocyte development and antigen signaling. CD45 isoforms arise via alternative splicing and differ in extracellular domains, affecting membrane localization and function. CD45 also modulates integrin-mediated signaling and apoptosis-related DNA fragmentation. CD45RA is a subtype with restricted expression among lymphoid cell types.
