Product Specific Information

The A20 monoclonal antibody reacts with the mouse CD45 molecule, also known as leukocyte common antigen (LCA), in CD45.1-expressing mouse strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed by all leukocytes in these strains.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; recommended ≤0.5 µg per test (100 µL final volume). Cell number may vary from 10^5 to 10^8 cells/test. Antibody titration recommended for optimal assay performance.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA-intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer Usage:

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails or single-color controls (10^3–10^8 cells per 100 µL sample).
• Contains formulation improvements over previous CellBlox buffer.

NovaFluor Technology:
Based on Phiton™ nucleic acid dye structures, engineered for minimal spillover and spread.
Excitation: 496 nm; Emission: 511 nm; Laser: 488 nm (Blue).

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, making up about 10% of lymphocyte surface proteins.
It plays a crucial role in lymphocyte development and antigen signaling by regulating Src-family kinases.
CD45 exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
Isoform expression varies by cell type and physiological state.
Additionally, CD45 contributes to integrin-mediated signaling, cell survival, and DNA fragmentation during apoptosis.
CD45RA is one isoform with restricted expression among lymphoid cell subtypes.
