Product Specific Information

The A20 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.1-expressing strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed by all leukocytes in these strains.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread effects.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells. It constitutes ~10% of lymphocyte surface proteins and plays a key role in lymphocyte development and antigen signaling by regulating Src-family kinases.

CD45 exists as multiple isoforms due to alternative splicing, differing in extracellular domains. Isoform expression varies by cell type and physiological state. CD45 also modulates integrin-mediated signal transduction and participates in DNA fragmentation during apoptosis. CD45RA is one isoform with restricted expression among lymphoid cell subtypes.
