Product Specific Information
This COX 229 monoclonal antibody recognizes human COX2 (PTGS2). It has been reported to cross-react with mouse and rat COX2. For optimal performance, use with the Foxp3 / Transcription Factor Staining Buffer Set and protocol.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on stimulated normal human peripheral blood cells using Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended ≤0.125 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.
Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis including inflammatory prostaglandins. The process involves:
- Cyclooxygenase (COX) reaction: arachidonate → prostaglandin G2 (PGG2)
- Peroxidase reaction: PGG2 → prostaglandin H2 (PGH2)
COX2 is constitutively expressed in endothelium, kidney, and brain, and up-regulated in pathological conditions such as cancer and inflammation. Overexpression is linked to increased cell adhesion, resistance to apoptosis, and tumor angiogenesis. COX2 mediates production of prostaglandin E2 (PGE2), influencing motility and proliferation. It is inhibited by calcitriol and glucocorticoids, and targeted by NSAIDs like aspirin.
