Product Specific Information
The M1/70 monoclonal antibody reacts with mouse CD11b (integrin alphaM, 165–170 kDa). CD11b associates with CD18 to form the Mac-1 (αMβ2 integrin) complex, binding to CD54 (ICAM-1), C3bi, and fibrinogen. Expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and peritoneal B-1 cells. Cross-reactive with human CD11b, allowing detection on human monocytes, granulocytes, and NK cell subsets.
Not recommended for use on formalin-fixed paraffin-embedded tissue.
Applications Reported: Flow cytometry and IHC (frozen tissue).
Applications Tested: Flow cytometric analysis of mouse splenocytes or bone marrow cells at ≤0.125 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize titration for best results.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule forming a heterodimer with CD18 (integrin beta-2). This complex, known as Mac-1 or CR3, mediates adhesion via ICAM-1, ICAM-2, ICAM-4, CD14, fibrinogen, and Factor X.
Expressed on monocytes/macrophages, granulocytes, NK cells, dendritic cells, and microglia. Expression increases during monocyte maturation and varies among tissue macrophages.
CD11b dysfunction is associated with systemic lupus erythematosus and ITGAM-related autoimmune susceptibility.
