Description:
The monoclonal antibody BCL-UP recognizes Bcl-6 (B-cell lymphoma 6), a member of the POZ family of transcriptional repressors. Bcl-6 is abundantly expressed in germinal center B cells and plays essential roles in B cell proliferation, survival, and differentiation. It is also expressed in diffuse large B cell lymphomas (DLBCLs), Burkitt’s lymphoma, and non-Hodgkin’s lymphoma, as well as in CD4+ T follicular helper and memory T cells. Bcl-6 is known to be antagonized by Blimp-1.

Applications Reported:
This antibody is suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular flow cytometry using human tonsil cells and the Foxp3 Fixation/Permeabilization Buffers (Product #00-5521-00).
Refer to BestProtocols® for staining protocol (Protocol B: One-step for intracellular (nuclear) proteins).
Use at 5 µL (0.03 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.

• Recommended filters: 685 LP dichroic mirror and 710/40 bandpass filter (695/40 acceptable alternative)

Stability:
PerCP-eFluor® 710 conjugated antibodies are stable when stained samples are exposed to 2% formaldehyde overnight at 4°C. Evaluate for alternative fixation protocols as needed.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Bcl-6 proto-oncogene product is a Kruppel-type zinc-finger protein expressed mainly in germinal center B cells and related lymphomas. It is involved in chromosome rearrangements at 3q27 in non-Hodgkin’s lymphomas, with rearrangements detected in 33–45% of diffuse large B cell lymphomas. Bcl-6 is also detected in follicular, diffuse large B cell, and Burkitt’s lymphomas, as well as in nodular lymphocyte predominant Hodgkin’s disease.
