Product Specific Information
For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments using this carrier-free antibody, perform an additional desalting step (recommended: Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest.
Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain. Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation.
p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer agent. It is also linked to aging, oxidative stress, and learning/memory processes. Normal cells express low levels, but transformed cell lines show high expression contributing to malignancy. Mutations in TP53 lead to loss of tumor suppressor activity and are found in various cancers and hereditary cancer syndromes such as Li-Fraumeni.
