Product Specific Information

The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.
In B cells, IRF4 is highly expressed in mature plasma cells and plays a crucial role in differentiation. It interacts with PU.1 and regulates transcription of B cell–specific genes such as Prdm1 (Blimp1).
In T cells, IRF4 is implicated in Treg, Th2, Th9, and Th17 cell development and function. It is upregulated upon T cell activation and induces Blimp1 expression in Tregs, leading to IL-10 expression.
IRF4 also contributes to macrophage polarization and regulation and has been reported to be upregulated in several hematologic malignancies.

Applications Reported:
This 3E4 antibody is validated for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells using the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00) and protocol.
Refer to BestProtocols® for staining intracellular antigens for flow cytometry (Protocol B: One-step protocol for intracellular (nuclear) proteins).
Use ≤0.5 µg per test in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test. Titrate antibody for optimal performance.

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure your instrument can detect this fluorochrome.
Laser: Violet Laser
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

The IRF (interferon regulatory factor) family of transcription factors features a unique tryptophan pentad repeat DNA-binding domain. IRFs regulate interferon responses to viral infection and control interferon-inducible genes.
IRF4 is lymphocyte-specific and negatively regulates toll-like receptor (TLR) signaling, essential for innate and adaptive immune activation.
A chromosomal translocation involving IRF4 and the IgH locus, t(6;14)(p25;q32), may cause multiple myeloma. Alternatively spliced transcript variants have been identified.
