Target Information

In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. Chromatin remodeling is a critical component of transcriptional regulation, and acetylation of nucleosomal histones plays a key role in this process. Acetylation of lysine residues in histone tails induces conformational changes that increase DNA accessibility to transcription factors.

Several mammalian proteins act as nuclear histone acetylases, including GCN5, PCAF (p300/CBP-associated factor), p300/CBP, HAT1, and TFIID subunit TAF II p250. Conversely, histone deacetylation is linked to transcriptional silencing. The histone deacetylase (HDAC) family includes HDAC1–9. HDAC9 and HDAC9a are alternatively spliced isoforms; HDAC9a is 132 amino acids shorter but retains catalytic activity and represses myocyte enhancer-binding factor 2-mediated transcription. Both isoforms are expressed in brain, skeletal muscle, kidney, placenta, and pancreas.
