This antibody is predicted to react with bovine, canine, mouse, and porcine based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is tonsil or Hodgkin’s lymphoma tissue.

Target Information

CD40 antigen is a single-chain glycoprotein belonging to the tumor necrosis factor/nerve growth factor superfamily. It shows significant homology to the Hodgkin’s disease-associated antigen CD30.
CD40 is expressed on all B cells except plasma cells, and also found on epithelial cells, carcinomas, and lymphoid dendritic cells.
It plays a crucial role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
The transcription factor AKNA coordinates CD40 expression, which may be important for homotypic cell interactions.
Adaptor protein TNFR2 interacts with CD40 to mediate signal transduction.
CD40-CD40L interaction is necessary for amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
