Target Information
RGMA is a member of the repulsive guidance molecule (RGM) family involved in multiple functions in the developing and adult nervous system. It regulates cephalic neural tube closure, inhibits neurite outgrowth and cortical neuron branching, and contributes to mature synapse formation.
RGMA binding to its receptor NEO1/neogenin activates the RHOA-ROCK1/Rho-kinase signaling pathway via the UNC5B-ARHGEF12/LARG-PTK2/FAK1 cascade, leading to neuronal growth cone collapse and neurite outgrowth inhibition.
Additionally, RGMA-NEO1 interaction influences HRAS-PTK2/FAK1-AKT1 signaling, resulting in HRAS inactivation.
RGMA also functions as a bone morphogenetic protein (BMP) coreceptor signaling through SMAD1, SMAD5, and SMAD8.
