HFE2는 철 대사에 관여하며, hepcidin 활성화 신호 경로의 구성 요소 또는 조절자로 작용할 수 있습니다. 또한 세포 내 hepcidin 수용체 역할을 할 가능성이 있습니다. 대체 스플라이싱된 전사체 변이체가 보고되었으며, 이 유전자의 결함은 hemochromatosis type 2A (juvenile hemochromatosis, JH)의 원인입니다. JH는 조기 발병하는 상염색체 열성 질환으로, 심각한 철 과부하로 인해 성선기능저하증, 간 섬유화 또는 간경변, 심근병증 등이 30세 이전에 발생할 수 있습니다.
