Target Information

RGMA is a member of the repulsive guidance molecule (RGM) family involved in multiple functions in the developing and adult nervous system. It regulates cephalic neural tube closure, inhibits neurite outgrowth and cortical neuron branching, and contributes to mature synapse formation.
Binding to its receptor NEO1/neogenin activates the RHOA-ROCK1/Rho-kinase signaling pathway through the UNC5B-ARHGEF12/LARG-PTK2/FAK1 cascade, leading to neuronal growth cone collapse and neurite inhibition.
Additionally, RGMA-NEO1 interaction affects HRAS inactivation via the HRAS-PTK2/FAK1-AKT1 pathway. RGMA also functions as a bone morphogenetic protein (BMP) coreceptor, signaling through SMAD1, SMAD5, and SMAD8.
