Target Information
Reticulon-1 (formerly designated NSP for Neuroendocrine-Specific-Protein) is specifically expressed in neural and neuroendocrine tissues.
The NSP gene is mapped to human chromosome 14q21–q22 and encodes three overlapping proteins: reticulon-1A (NSP-A), reticulon-1B (NSP-B), and reticulon-1C (NSP-C).
These proteins are anchored to endoplasmic reticulum membranes via their common C-terminal regions.
- Reticulon-1A: ~135 kDa, occurs in multiple isoforms depending on phosphorylation; a reliable marker for neuroendocrine differentiation in lung cancer diagnosis (SCLC and carcinoid tumors).
- Reticulon-1B: ~45 kDa phosphoprotein, restricted to lung cancer cell line NCI-H82, not found in human tissues.
- Reticulon-1C: ~23 kDa non-phosphorylated protein, found with Reticulon-1A in SCLC cell lines but not in non-SCLC cultures.
