Product Specific Information

Description:
The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2) and serves as a potent co-stimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells.

Applications Reported:
For use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometry of normal human peripheral blood cells. Use at 5 µL (1.0 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Violet™ 650 (BV650) is a tandem dye emitting at 649 nm, optimized for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
BV650 is sensitive to photo-induced oxidation. Protect from light during storage and staining.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark. BV650 is not compatible with methanol-based fixation.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T-cell specific surface glycoprotein belonging to the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and provides the crucial co-stimulatory signal for T-cell activation by binding CD80 and CD86 on antigen-presenting cells. CD28 prevents T-cell anergy and premature apoptosis. Expression patterns vary across thymocyte maturation stages. CD28 dysfunction has been associated with mycosis fungoides and Sezary’s disease.
