Product Specific Information

The LG.7F9 monoclonal antibody reacts with mouse CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by virtually all mature T cells and a subpopulation of B cells (mainly memory B cells). In mouse, CD27 is found on nearly all thymocytes except CD46-CD8- precursors. CD27 binds CD70 and plays an important role in T cell–B cell interaction and effector cell maturation. LG.7F9 cross-reacts with human and rat CD27.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; ≤1.0 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimal titration recommended.

Brilliant Violet™ 650 (BV650) emits at 649 nm and is intended for use on cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54).
BV650 is not compatible with methanol-based fixation.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily (including CD40, CD30). It regulates cell proliferation and death. CD27/CD70 expression is confined to lymphocytes, with CD27 highly expressed upon antigen receptor ligation and CD70 requiring co-stimulatory signals. CD27 forms a disulfide-linked homodimer on thymocytes, peripheral T cells, and memory B cells. Activation via TCR-CD3 increases CD27 expression and releases soluble CD27 (sCD27, 28–32 kDa), a prognostic marker for B cell malignancies. CD27-binding protein (SIVA) mediates apoptosis. CD27 dysfunction is associated with Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
