Diacylglycerol kinases (DGKs) phosphorylate diacylglycerol (DAG) to produce phosphatidic acid. DAG and phosphatidic acid act as second messengers in signaling cascades.
DGK subtypes exhibit tissue-specific expression and functional diversity:
- DGK-α: Influences cell activation and exosome secretion
- DGK-β: Abundant in caudate putamen and olfactory tubercle
- DGK-γ: Expressed in retina and other tissues
- DGK-δ: Most abundant in skeletal muscle
- DGK-ε: Specific for arachidonyl-DAG, predominant in testis
- DGK-θ: Abundant in cerebellum and hippocampus
- DGK-ι: Expressed in brain and retina
- DGK-η: Closely related to DGK-δ
- DGK-ζ: Abundant in brain and muscle
