Product Specific Information

The 2H7 monoclonal antibody reacts with human CD20, a 33–36 kDa transmembrane protein expressed by developing and mature B cells, but not plasma cells. CD20 may play a role in B-cell activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Super Bright 780 is a tandem dye excited by a 405 nm violet laser and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

For compensation setup, use cells rather than beads due to higher compensation values observed with UltraComp eBeads ([Product # 01-2222-42]) and AbC Total Antibody Compensation beads ([Product # A10497]).

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer ([Product # 00-8222]) or 1-step Fix/Lyse Solution ([Product # 00-5333]) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 remains on the cell surface after antibody binding, making it an effective target for therapeutic B-cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
