Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early T and B progenitors and activated mature lymphocytes. It binds IL-2 with low affinity alone but forms a high-affinity receptor complex with CD122 and CD132. Binding of IL-2 to both receptor classes is inhibited by PC61.5.

Fluorochrome Information:
Super Bright 780 is a tandem dye excited at 405 nm and emits at 780 nm. Use a 780/60 bandpass filter. Protect from light due to photo-induced oxidation sensitivity.

Staining Recommendations:
Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright-conjugated antibodies to minimize non-specific polymer interactions.

Compensation Notes:
Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42) compared to stained cells. In such cases, set compensation with cells. Similar results observed with AbC Total Antibody Compensation beads (Product # A10497).

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal brightness loss.

Target Information

CD25 (IL2RA) encodes the IL-2 receptor alpha chain, part of the heterotrimeric IL-2 receptor complex (with IL2RB and IL2RG). It regulates T and B lymphocyte proliferation and immune response. IL-2RA homodimers form low-affinity receptors; IL-2RB homodimers form medium-affinity receptors. IL-2 is mainly produced by mature T cells and functions as a growth and differentiation factor. Mutations in IL2RA are associated with immune dysregulation diseases such as ulcerative colitis-like conditions in mice.
