Product Specific Information

Description:
The N418 monoclonal antibody reacts with mouse CD11c (integrin alphaX), which associates with beta2 integrin to form the CD11c/CD18 heterodimer. CD11c is expressed by dendritic cells, intestinal intraepithelial lymphocytes, and some activated T cells. It binds CD54, iC3b, and fibrinogen, playing a role in leukocyte adhesion. N418 binds CD11c on splenic dendritic cells and precipitates a 150/90 kDa heterodimer.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg/test in 100 µL final volume. Cell number may vary (10⁵–10⁸ cells/test). Optimize concentration for specific assays.

Fluorochrome Information:
Super Bright 780 is excited by 405 nm (violet laser) and emits at 780 nm. Recommended filter: 780/60 bandpass or equivalent. Verify instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to reduce non-specific interactions.

Compensation Notes:
Higher compensation values may occur in violet 450/50 channel with UltraComp eBeads ([Product # 01-2222-42]) or AbC Total Antibody Compensation beads ([Product # A10497]). Prefer cell-based compensation.

Light Sensitivity:
Protect from light; dye is photo-sensitive.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark. Clone-specific performance should be empirically verified.

Target Information

CD11 (integrin alpha chain) forms a heterodimer with CD18, functioning as an adhesion molecule.

• CD11a: Expressed in B/T lymphocytes, macrophages, monocytes, neutrophils, basophils, eosinophils
• CD11b: Expressed in monocytes, macrophages, NK cells, some B/T cells, granulocytes
• CD11c: Expressed in monocytes, macrophages, NK cells, some granulocytes, and subsets of lymphocytes
