Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM that associates with CD18 to form the alphaMbeta2 integrin (Mac-1). It binds to CD54 (ICAM-1), C3bi, and fibrinogen. CD11b is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and mouse B-1 cells. M1/70 cross-reacts with human CD11b, allowing detection on human monocytes, granulocytes, and NK cells.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Validated on mouse bone marrow cells at ≤0.125 µg/test (100 µL final volume). Optimal antibody concentration should be titrated experimentally.

Super Bright 780 is a tandem dye excited by the 405 nm violet laser and emits at 780 nm. Use a 780/60 bandpass filter. Verify instrument compatibility.

When using multiple Super Bright conjugates, apply Super Bright Complete Staining Buffer (Product # SB-4401) to reduce non-specific interactions.
For compensation setup, use stained cells rather than beads if higher compensation values are observed.

Light Sensitivity: Protect vial and samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer or 1-step Fix/Lyse Solution with minimal impact on brightness.

Target Information

CD11b (integrin alpha-M, ITGAM) forms a heterodimer with CD18 known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, lymphocytes, NK cells, dendritic cells, and microglia. CD11b/CD18 mediates adhesion via ICAMs, CD14, CD50, CD23, fibrinogen, and Factor X. It serves as a microglial marker and varies by tissue macrophage type. Dysfunctions are linked to systemic lupus erythematosus and ITGAM-related susceptibility.
