Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subsets of CD4+ and CD8+ peripheral T lymphocytes and B cells. CD45RA+ T cell populations are mainly naive/virgin, allowing the use of HI100 mAb as a phenotypic marker to discriminate T cell subsets.

Fluorochrome Information:
Brilliant Ultra Violet™ 496 (BUV496) emits at 496 nm and is intended for use on cytometers with an ultraviolet (355 nm) laser. Ensure the instrument supports this fluorochrome.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and compensation.

Target Information

CD45RA is an isoform of the CD45 complex, expressed in specific lymphoid subtypes. CD45 (Leukocyte Common Antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases, critical for lymphocyte development and antigen signaling.

CD45 isoforms differ by alternative splicing of extracellular domains, influencing membrane localization and cell-type expression. Beyond immunoreceptor signaling, CD45 modulates integrin-mediated pathways and apoptosis-related DNA fragmentation. It is expressed on all human hematopoietic lineage cells except mature red cells.
