Description:
The 104 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.2-expressing mouse strains such as C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.
Applications Reported:
This antibody is reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1.0 µg per test (100 µL final volume). Optimal concentration should be titrated by user.
Fluorochrome Information:
Brilliant Ultra Violet™ 496 (BUV496) emits at 496 nm and is intended for use with ultraviolet (355 nm) laser-equipped cytometers. Verify instrument compatibility before use.
Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.
Light Sensitivity:
Protect from light to prevent photo-induced oxidation.
Fixation:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-Step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark.

Target Information
CD45.2 (LCA) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases and participates in lymphocyte development and antigen signaling. Isoforms arise from alternative splicing, differing in extracellular domains. CD45.2 also modulates integrin-mediated signaling and apoptosis-related DNA fragmentation. CD45RA is an isoform with restricted expression among lymphoid subtypes.
