Target Information
This gene encodes a G protein-coupled receptor selective for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant). These chemokines belong to the CXC subfamily, characterized by a single amino acid separating the first two conserved cysteines.
Binding of chemokines to CXCR3 induces cellular responses involved in leukocyte trafficking, integrin activation, cytoskeletal rearrangement, and chemotaxis. The receptor is coupled to Gi-type G proteins, and signal transduction likely involves enzymes shared with other chemokine receptor pathways.
CXCR3 expression is prominent in effector/memory T cells and T cells from inflamed tissues. IP10, Mig, and I-TAC are often produced locally in inflammatory lesions, implicating CXCR3 in inflammatory cell recruitment. Thus, CXCR3 is a therapeutic target for small-molecule antagonists aimed at treating inflammatory diseases. Multiple transcript variants encoding different isoforms have been identified.
